Fibers keep cilia regular

چکیده
Fibers keep cilia regular A ctin fi bers and microtubules help cilia coordinate the direction and sequence of their beating, Werner et al. show. At fi rst, the multiple cilia on an embryonic cell are a bit like the musicians in a garage band, each doing its own thing. But over time the fi laments turn so that they all beat in the same direction. They also coordinate their timing, so that the cilia at the front of the cell beat fi rst and the ones at the back of the cell stroke last. The planar cell polarity signaling pathway and hydrodynamic forces on the cilia help set up this polarity. Using the cilia-coated epithelial cells of Xenopus embryos, Werner et al. investigated the roles of the actin and microtubule cytoskeletons in the process. Werner et al. observed actin fi bers connecting neighboring cilia. These fi laments were absent in the youngest cells that hadn’t yet polarized. To determine the fi bers’ function, the researchers disassembled them using the drug cytochalasin D. Without actin fi bers, the cilia couldn’t reorient to beat in the same direction. They also couldn’t coordinate their timing. However, neighboring clusters of cilia could still synchronize. This local coordination disappeared in cells lacking microtubules, the team found. These results indicate that two components of the cytoskeleton perform different functions: actin fi bers establish cell-wide coordination of cilia orientation and timing, whereas microtubules ensure that cilia are in harmony with their neighbors. An Effect of Adenosine Triphosphate on the Light Scattered by Suspensions of Cilia
Cilia were isolated from Tetrahymena pyriformis by an ethanol-calcium method. Addition of adenosine triphosphate to a suspension of intact or digitonin-extracted cilia caused a decrease of about 20 per cent in turbidity. Study of fractionated cilia showed that the presence of two distinct axonemal components, the outer fibers and the 30S dynein (the axonemal ATPase protein), was necessary for t...
متن کاملThe Relationship between the Fine Structure and Direction of Beat in Gill Cilia of a Lamellibranch Mollusc
This paper describes the fine structure and its relationship to the direction of beat in four types of cilia on the gill of the fresh-water mussel Anodonta cataracta. The cilia contain nine outer, nine secondary, and two central fibers, such as have been described previously in other material. Each outer fiber is a doublet with one subfiber bearing arms. One particular pair of outer fibers (num...
متن کاملUltrastructural Organization of Cilia and Basal Bodies of the Epithelium of the Choroid Plexus in the Chick Embryo
Ultrastructural studies were performed on normal and abnormal cilia and basal bodies associated with the choroidal epithelium of the chick embryo. Tissues were prepared in each of several fixatives including: 1% osmium tetroxide, in both phosphate and veronal acetate buffers; 2% glutaraldehyde, followed by postfixation in osmium tetroxide; 1% potassium permanganate in veronal acetate buffer. No...
متن کاملCell Motility: Deaf Drosophila Keep the Beat
Intraflagellar transport is involved in the assembly of cilia and eukaryotic flagella. Two recent studies have shown that defects in intraflagellar transport prevent assembly of sensory cilia in Drosophila, leaving the fly deaf and uncoordinated. Surprisingly, the mutant sperm flagella have normal structure and motility.
متن کاملOlfactory Cilia in the Frog
Olfactory epithelium from the frog was examined in the living state by light microscopy and in the fixed state by electron microscopy. Particular attention was paid to the layer of cilia and mucus which covers the surface of the epithelium. The olfactory cilia differed from typical cilia in that they (a) arose from bipolar neurons and had centrioles near their basal bodies, (b) were up to 200 m...
